New water-soluble fluorescent tetracationic imidazolium-based macrocycles are synthesized via a modular S_N2 nucleophilic substitution reaction. The positive charge and acidic C–H sites of these macrocycles enable them to bind with nucleotides in water, driven by hydrogen bonds and electrostatic interactions. The binding is high affinity for suitable nucleotides. These properties position them as promising candidates for the selective sensing of nucleotides.
