摘要: 建立了稳定同位素稀释-液相色谱-串联质谱法快速检测血浆和尿液中米酵菌酸和异米酵菌酸的方法。样品经乙腈蛋白沉淀净化、反相C18色谱柱分离后,采用电喷雾串联质谱,选择反应监测(SRM)模式下检测,溶剂标准曲线内标法定量。在2.0~400μg/L浓度范围内,米酵菌酸和异米酵菌酸呈良好的线性关系,相关系数大于0.9984;方法定量限(10倍基线噪声对应的浓度)为2.0μg/L; 2.0、10.0、40.0和200.0μg/L的加标水平下,方法的日内和日间回收率分别为89.7%~109.6%和86.2%~104.2%,日内与日间相对标准偏差均小于9.8%。本方法简单、快速、灵敏、特异性强,可满足人体生物样本中米酵菌酸和异米酵菌酸的公共卫生应急检测或临床诊断检测要求。
