金黄色葡萄球菌候选特征肽的靶向分析

摘要: 采用液相色谱-串联质谱联用的靶向分析技术,通过检测食源性致病菌金黄色葡萄球菌水解生成的特征肽,实现食品中致病菌的高效检测。针对文献报道的金黄色葡萄球菌的9种候选特征肽,验证了这些特征肽的靶向分析方法,考察和比较了分析效能。结果表明,基于多反应监测模式的靶向分析方法灵敏度高,更适于特征肽的检测;确定了每个特征肽最佳的碎片离子质量,进一步的生物样品分析结果表明,I1(AYLAVPAAPK)、I2(DYNSPTLIGWVK)、I5(AGYTVNNTPK)和I6(SISSGYTSGR)更适合作为金黄色葡萄球菌的候选特征肽。本研究为大规模验证候选特征肽的鉴定效果奠定了基础,为特征肽检测策略在食源性致病菌检测中的应用提供了参考。
